AbstractThis introductory Laboratory course introduces the student to the entire range of classical and modern molecular biosciences. In the second year (Praktikum GL Bio II) the students will perform three praktikum days in:
- Bioinformatics
- Plant Physiology
- Genetics and
- Cell Biology II.

(total of 12 experiments)

Each experiment takes one full day.

ContentThe class is divided into four blocks: Cell Biology II, Genetics, Bioinformatics and Plant Physiology. One block lasts three weeks.

CELL BIOLOGY II:
- Cells: Cell types & staining methods, cell fusion & cell motility,
- Tissue and development: histology of mice embryos & embryogenesis
- Repair mechanisms: DNA repair & wound healing,

GENETICS:
- Yeast genetics
- Drosophila genetics
- Human genetics

BIOINFORMATICS:
- the command line interface (UNIX/LINUX)
- reinforce programming skills in R
- programmatic, analytical and statistical principles

PLANT PHYSIOLOGY:
- Plants and light
- Phytohormones and other growth factors
- Molecular biology of systemic gene silencing
- Literature and presentations

The students will also prepare short presentations (approx. 10 min) of the various topics within this course.

Prerequisites / NoticeTHE PRAKTIKUM RULES:

Your attendance is obligatory and you have to attend all 12 Praktikum days. Absences are only acceptable if you are able to provide a Doctor’s certificate. The original Dr's certificate has to be given to Dr. M. Gstaiger (HPM F43) within five days of the absence of the Praktikum day.

If there will be any exceptional or important situations then you should directly contact the Director of Studies of D-Biol, who will decide if you are allowed to miss a Praktikum day or not.

HIGHLY IMPORTANT!!

1. Due to the increased number of students, the official Praktikum registration has to be done, using myStudies, preferably at the end of HS20 but not later than Wednesday January 29, 2021.

2. Later registration is NOT possible and can NOT be accepted!

3. The course registration for FS21 is usually possible at the end of HS20 and you will obtain an E-mail from the Rectorate when the course registration using myStudies is possible.

Students can register for a practice group via myStudies. As soon as the course unit is registered in myStudies, a text box appears indicating that a group can be selected. Accordingly, students can select a group in the next step. If more than 180 students register, the surplus students will be placed on a waiting list and then allocated by the course responsible.


The Praktikum GL BioII FS21 will take place during the following days and therefore, you have to make sure already now that you do not have any other activities & commitments during these days:

AbstractThis 1st year Laboratory course introduces the students to the basic experimental techniques used in the classical and modern molecular biosciences. In the first year (Praktikum GL BioI) the students will be introduced into the basic concept and methods of Microbiology, Biochemistry and Molecular Biology in 12 consecutive course days.
ObjectiveIntroduction to biology with practical work.

ContentThis 1st year Laboratory course introduces the student to the entire range of classical and modern molecular biosciences. Students will participate in 12 consecutive course days.

Because of necessary Covid-19 safety measures the practical will operate in two shifts to reduce the number of student in the practical rooms.

1. shift 8:00-13:00
2. shift 13:30-18:30

Program

Day 1: Isolation and cultivation of microorganisms
Day 2: Morphological, biochemical and genetic characterization of microorganisms
Day 3: Evolution of microorganisms and their abiotic and biotic interactions
Day 4: Lifestyle of fungi as eukaryotic representatives of the microorganisms
Day 5: DNA extraction, tranformation (E.coli, yeast)
Day 6: RNA extraction, Life cycle of eukaryotic cells
Day 7: Purification of TAQ polymerase from E coli by ionexchange chromatography
Day 8: Characterizing fractions of the purification of TAQ polymerase (SDS-PAGE, WB)
Day 9: mRNA splicing in yeast by RT-PCR and purified TAQ polymerase
Day 10: Affinity chromatography, protein crystallization and structure
Day 11: Protein folding, protein stability and Enzyme kinetics
Day 12: Protein folding, protein stability and Enzyme kinetics

AbstractApplication of current experimental strategies to study the dynamics of complex and highly regulated cellular processes.
ObjectiveIn this course, students will
- learn what principles govern cellular dynamics and how these are regulated.
- learn to evaluate and to apply current strategies to study the dynamics of complex and highly regulated cellular processes
ContentDuring this Block-Course, the students will learn to
(1) describe the important mechanisms and regulators of dynamic processes in cells,
(2) perform experimental techniques to quantify dynamic cellular processes,
(3) evaluate and compare experimental strategies and model systems,
(4) formulate and present scientific concepts in an oral presentation.

Topics discussed will include
- mobility in the cell (passive and active)
- compartmentalization (by membranes and via phase separation)
- examples of cell biological processes dependent on mobility and compartmentalization.

Students will work in small groups in individual labs on one research project (8 full days of practical work; every group of students will stay in the same lab during the entire course). The projects are close to the actual research carried out in the participating research groups, but with a clear connection to the subject of the course.
LiteratureDocumentation and recommended literature (review articles and selected primary literature) will be provided during the course.
